A mate of mines got some ET28 Speedlines for his Corsa, however, will he have to reduce the back of the rim to 49 in a lathe OR get spacers for them??

Im sure its a lathe jobbie?

Spacers LOWER the offset.
So you will need to reduce the back of the rim.

Its likely you wont be able get them to et49 though, it will weaken the wheel too much.
Most engineering places that do the sort of thing will say about 5mm max

Sh1tter, they need to go down like 24mm!!! To get to 49.

lol hes funked then lol Even a take down to ET38 is a good 10mm!

I've asked about skimming wheels before and most people said not to take any more than 4-5mm off! So he is screwed! :)
